摘要: The flavivirus capsid protein not only is a component of nucleocapsids but also plays role in viral replication. In this study, we found small cells infected with Japanese encephalitis virus (JEV) the particles. was shown to be generated by processing host cysteine protease cathepsin L. An vitro cleavage assay revealed that L cleaves between amino acid residues Lys 18 and Arg 19 , which are well conserved among mosquito-borne flaviviruses. A mutant JEV resistant from an infectious cDNA clone introducing substitution site. exhibited growth kinetics similar those wild-type monkey (Vero), mosquito (C6/36), porcine (PK15) cell lines, whereas replication mouse macrophage (RAW264.7) neuroblastoma (N18) impaired. Furthermore, neurovirulence neuroinvasiveness mice were lower than JEV. These results suggest crucial neural cells, leads pathogenesis infection.
