摘要: We have investigated human gliomas that amplify and rearrange the epidermal growth factor receptor gene, with generation of an in-frame deletion mutation 802 nucleotides in external domain. This generates a local amino acid sequence at fusion junction what normally were distant polypeptide sequences intact receptor. 14-amino peptide was chemically synthesized, coupled to keyhole limpet hemocyanin, used as immunogen rabbits. The elicited antibody reacted specifically ELISA. anti-fusion purified by passage antiserum over affinity column acidic elution. selectively bound glioma mutant compared assessed immunocytochemistry, immunofluorescence, immunoprecipitation gel electrophoresis, binding experiments using radioiodinated antibody. These data indicate it is feasible generate site-specific anti-peptide antibodies are highly selective for proteins tumors. described here, other antibodies, should be ideal candidates tumor immunoimaging immunotherapy.
