Gene expression profiling of bovine endometrium during the oestrous cycle: detection of molecular pathways involved in functional changes.

摘要: The endometrium plays a central role among the reproductive tissues in context of early embryo–maternal communication and pregnancy. It undergoes typical changes during sexual/oestrous cycle, which are regulated by ovarian hormones progesterone oestrogen. To identify underlying molecular mechanisms we have performed first holistic screen transcriptome bovine intercaruncular at two stages cycle – end day 0 (late oestrus, low progesterone) 12 (dioestrus, high progesterone). A combination subtracted cDNA libraries array hybridisation revealed 133 genes showing least 2-fold change their mRNA abundance, 65 with higher levels oestrus 68 dioestrus. Interestingly, were identified showed differential expression between different uterine sections as well. most prominent example was UTMP (uterine milk protein) mRNA, markedly upregulated cranial part ipsilateral horn oestrus. Gene Ontology classification known function characterised time elevated genes, for related to cell adhesion, motility extracellular matrix dioestrus mRNAs encoding variety enzymes transport proteins, particular ion channels. Searching pathway databases literature data-mining physiological processes signalling cascades, e.g. transforming growth factor- retinoic acid signalling, potentially involved regulation oestrous cycle.
