Cross-reactivity between MUC1 antigen and MCA: false elevation of serum CA 15-3 level in pregnant and lactating women by Ma695-Ma552-based assay.

摘要: Cancer antigen 153 (CA 15-3) is one of the most commonly used biomarkers breast cancer. However, elevated CA 15-3 reported in pregnant and lactating women more frequently on Beckman DxI 800 immunoassay system (Ma695–Ma552 antibody pair) than Abbott ARCHITECT (115D8–DF3 laboratory methodological evaluation. We conducted this study order to figure out reason behind phenomenon. Serum concentration was analyzed 426 subjects, including 180 patients with cancer, 121 benign disease, 125 healthy volunteers (45 80 non-pregnant women). assay further validated using another cohort 112 or postpartum women. Immunological cross reaction by Western blotting immunoprecipitation. The serum level abnormally higher almost 95% detected Ma695–Ma552 pair (median: 71.4 U/mL) that 115D8–DF3 16.5 U/mL). immunoprecipitation indicated such a significant difference mainly due between monoclonal Ma552 mucin-like carcinoma-associated (MCA). suitable for monitoring therapy pregnancy-associated
