HER-2/neu oncogene amplification in stage I and stage III ovarian papillary serous carcinoma

摘要: Abstract Oncogene amplification has been implicated in the genesis and progression of many cancers. Overexpression HER-2/neu proto-oncogene occurs 20–30% ovarian epithelial cancers, which it may be prognostic significance. overexpression is traditionally studied using immunohistochemistry. In this study we used fluorescent situ hybridization (FISH) to determine papillary serous carcinoma compared frequency two stages disease. Archival tissues from 23 cases (9 stage I 14 III) were analyzed by FISH a probe chromosome 17 centromere control probe. Determination level was performed according standard protocols Cytogenetics Laboratory at Rhode Island Hospital. Of successfully analyzed, among tumors 22% (2/9) III 71% (10/14). These results are significant ( P = 0.036). The amplified 17% (27/12) 83% (10/12). This not only confirms presence subset with gene amplification, but also indicates that oncogene more likely associated advanced stage. Thus, present data consistent hypothesis similar protein over expression, marker poor outcome.
