摘要: Endothelial cells represent important targets for therapeutic and diagnostic interventions in many cardiovascular, pulmonary, neurological, inflammatory, metabolic diseases. Targeted delivery of drugs (especially potent labile biotherapeutics that require specific subcellular addressing) imaging probes to endothelium holds promise improve management these maladies. In order achieve this goal, drug cargoes or their carriers including liposomes polymeric nanoparticles are chemically conjugated fused using recombinant techniques with affinity ligands endothelial surface molecules. Cell adhesion molecules, constitutively expressed on the exposed pathologically altered endothelium—selectins, VCAM-1, PECAM-1, ICAM-1—represent good determinants such a delivery. particular, PECAM-1 ICAM-1 meet criteria accessibility, safety, relevance (patho)physiological context treatment inflammation, ischemia, thrombosis offer unique combination targeting options anchoring as well intra- transcellular targeting, modulated by parameters design system local biological factors flow phenotype. This review includes analysis examples selected classes therapeutics showing promising results animal studies, supporting translational potential interventions.
