摘要: Several physiological and pathophysiological events involving vascular endothelium occur at the microvascular level. Studies on human microvasculature require homogenous primary cultures of endothelial cells. However, procedures available for isolating culturing dermal cells (HDMEC) result in significant contamination with fibroblasts. To eliminate fibroblasts or other cells, we developed a procedure to isolate HDMEC from neonatal foreskin by panning using EN4, an anti-endothelial cell monoclonal antibody. Panned uniformly expressed von Willebrand factor CD36, confirming their characteristics, whereas cultured without showed degree In presence growth (VEGF), could be under serum-free conditions. VEGF stimulated dose-dependent manner medium media supplemented either serum newborn calf serum. Since differences exist between large vessel compared response stimulation umbilical vein (HUVEC). The dose two types was different. This effect may mediated receptor kdr, since mRNA kdr detected RT-PCR both HUVEC. described this study will make possible culture highly enriched facilitate studies these defined assay conditions environment.
