Simultaneous detection of X‐ and Y‐bearing human sperm by double fluorescence in situ hybridization

摘要: Double fluorescence in situ hybridization (FISH) was used to detect sex chromosomes decondensed human sperm nuclei. Biotinylated X chromosome specific (TRX) and digoxigenin-labeled Y (HRY) probes were simultaneously hybridized preparations from 12 normal healthy donors. After the hybridization, detected immuno-cytochemically, using two different independent affinity systems. Ninety-six percent of 12,636 showed fluorescent labeling, which 47.4% haploid 46.8% Y. A frequency 0.46% XX-bearing (0.28% disomic, 0.18% diploid) 0.38% YY-bearing (0.21% 0.17% found. The overall proportions X- Y-bearing ejaculates 47.9% 47.2%, respectively, not significantly expected 50:50 ratio. In addition 0.21% cells appeared be XY-bearing sperm, 0.62% diploid cells, 0.05% considered tetraploid cells. application double FISH X-chro-mosome Y-chromosome has allowed a more accurate assessment chromosal complements than single method quinacrine staining for Y-bodies. © 1993 Wiley-Liss, Inc.
