Mycobacterium tuberculosis DNA detection in soluble fraction of urine from pulmonary tuberculosis patients.

摘要: CONTEXTE : Une institution de soins tertiaires et recherche en Italie. OBJECTIF: De petits fragments l'ADN provenant cellules mortes au sein l'organisme ont ete detectes dans l'urine (ADN transrenal, ADN-Tr). Nous avons vise a tester l'hypothese que le ADN-Tr Mycobacterium tuberculosis pourrait etre detecte des patients atteints tuberculose (TB) pulmonaire. SCHEMA: etudie 43 dont la TB pulmonaire avait confirmee par culture ou n'existait aucun signe d'atteinte extra-pulmonaire, 10 maladies pulmonaires autres 13 sujets-controle bien portants. On extrait on l'a analysee une polymerase chain reaction (PCR) « semi-nested ». RESULTATS: trouve sequences specifiques M. 34 (79%) tuberculeux etudies, alors negative chez tous les sujets-controle. La nature transrenale demontree deux types d'evidence: premierement, l'analyse separee surnageants sediments huit etudies montre sept positifs seulement apparies. Deuxiemement, amplifiees PCR avec amorces destinees amplicons courts mais non longs. CONCLUSION: peut detecter d'une proportion significative d'ADN tuberculosis. Si ces observations sont confirmees etudes plus importantes, technologie representer nouvelle approche detection l'infection
