摘要: Neurofibromatosis type 1 (NF1) is one of the most common human genetic diseases affecting nervous system and predisposes individuals to cancer, including peripheral nerve sheath tumors (PNSTs) astrocytomas. Modifiers in background affect severity disease we have previously mapped two modifier loci, Nstr1 Nstr2, that influence resistance PNSTs Nf1−/+;Trp53−/+cis mouse model NF1. We report here analysis isolation from other epistatic loci using a chromosome substitution strain, further show locus (or loci) on 19 influences both This interacts with sex, resulting sex-specific modification tumors. Allele variability affects timing penetrance growth different tumor types associated NF1, specifically astrocytoma. These results indicate modifiers cancer susceptibility interact tumorigenesis under conditions demonstrate power strains study modifiers.
