摘要: Objective To understand the mechanism underlying gestational-onset hypothyroidism–induced male infertility. Design Controlled laboratory study. Setting Research in a university department of endocrinology. Animal(s) Wistar rat. Intervention(s) Pregnant rats were exposed to methimazole from embryonic days 9 14, 18, and 21, covering specific fetal periods differentiation development reproductive tract organs. Main Outcome Measure(s) Fertility was assessed by testing sperm count, forward motility, vivo fertilizing ability. Secretory activity epididymis evaluated quantifying sialic acid, carnitine, glycerylphosphorylcholine. Bioavailability androgens testosterone serum testicular interstitial fluid epididymal 5α-reductase activity/mRNA expression. Androgen receptor (AR) status tested detecting expression levels its mRNA protein, as well ligand binding activity. Data analyzed statistically one-way analysis variance. Result(s) Gestational exposure decreased ability, bioavailability androgens, AR status, secretory adult rats. Conclusion(s) Transient gestational - onset hypothyroidism affects fertility impairing posttesticular maturation process epididymis, owing subnormal androgen(s) bioavailability, expression, functional
