Modulation of epidermal growth factor receptor gene transcription by a polymorphic dinucleotide repeat in intron 1.

摘要: The influence of a highly polymorphic CA dinucleotide repeat in the epidermal growth factor receptor (EGFR) gene on transcription was examined with quantitative nuclear run-off method. We could demonstrate that EGFR is inhibited by approximately 80% alleles 21 repeats. In experiments polymerase chain reaction products spanned region more than 4,000 base pairs and contained promoter, two enhancers, first intron gene, we found activity declines increasing numbers dinucleotides. vivo pre-mRNA expression data from cultured cell lines support these findings, although other regulation mechanisms can outweigh this effect. addition, showed under our experimental conditions RNA elongation terminates at site closely downstream simple sequence there are separate major start sites. Our results provide new insights individually different role proto-oncogene.
