Systematic Review on the Involvement of the Kynurenine Pathway in Stroke: Pre-clinical and Clinical Evidence.

摘要: Background: Stroke is the second leading cause of death after ischemic heart disease and third disability-adjusted life-years lost worldwide. There a great need for developing more effective strategies to treat stroke its resulting impairments. Among several neuroprotective tested so far, kynurenine pathway (KP) seems be promising, but evidence still sparse. Methods: Here, we performed systematic review preclinical clinical studies evaluating involvement KP in stroke. We searched keywords: ("kynurenine" or "kynurenic acid" "quinolinic acid") AND ("ischemia" "stroke" "occlusion) electronic databases PubMed, Scopus, Embase. A total 1,130 papers was initially retrieved. Results: After careful screening, forty-five were included this review, being 39 pre-clinical six studies. Despite different experimental models cerebral ischemia, results are concordant implicating pathophysiology Preclinical also suggests that treatment with KMO inhibitors decrease infarct size improve behavioral cognitive outcomes. Few have investigated human stroke, consistent findings activated Conclusion: Well-designed addressing expression enzymes metabolites specific cell types their potential effects at cellular levels alongside warranted confirm translational as pharmacological target related complications.
