Protein overexpression and gene amplification of HER-2 and EGFR in colorectal cancers: an immunohistochemical and fluorescent in situ hybridization study

摘要: Overexpression of HER-2 and the epidermal growth factor receptor (EGFR) has been observed in many cancers, sometimes accompanied by gene amplification. To assess whether novel chemotherapies targeting these overexpressed proteins may be effective for treatment colorectal we examined exact frequency EGFR overexpression, relationship between amplification protein expression, heterogeneity within primary metastatic tumors. We evaluated 244 cancers immunohistochemically. All tumors found to overexpress or were further analyzed fluorescent situ DNA hybridization. was 8 (3%) 19 (8%) carcinomas, respectively. Gene 100 58% exhibiting cancer cells characterized clusters hybridization signals, suggesting amplicons homogeneously staining regions that predominant most amplification, as scattered signals reminiscent double minute chromosomes, coamplification with centromeric a minor population tumors, variety populations only small fraction but frequently
