摘要: BACKGROUND The down-regulation of the estrogen receptor-β (ERβ) gene is associated with several malignancies, including prostate carcinoma. The purpose current study was to investigate mechanisms ERβ inactivation through analysis CpG methylation promoter region gene. METHODS ERβ protein expression examined by immunohistochemistry in 23 cases human carcinoma and 40 benign prostatic hyperplasia (BPH). DNA extracted from these tissues processed for sodium bisulfite genomic sequencing. percentage sites (−376 −117), which contains 19 sites, determined sequencing data. cell lines DU145 ND1 were treated demethylating agent 5-AZAC mRNA analyzed reverse transcriptase-polymerase chain reaction. RESULTS In BPH tissues, found mainly epithelial cells. lacking 83% samples (19 samples) whereas all (40 40) demonstrated protein. mechanism because degree at within between −376 −117 higher compared tissues. Nine displayed significant differences samples. appeared lack expression. However, treatment restored those lines, suggesting that inactivates carcinoma. CONCLUSIONS The results demonstrate, what we believe be first time, occurs this gene. Cancer 2001;92:2076–83. © 2001 American Society.
