Small cell and non small cell lung cancer form metastasis on cellular 4D lung model

摘要: Metastasis is the main cause of death for lung cancer patients. The ex vivo 4D acellular model has been shown to mimic this metastatic process. However, concern model’s lack cellular components tumor’s microenvironment. In study, we aim determine if intact microenvironment will still allow metastasis form. We harvested a heart-lung block from rat and placed it in bioreactor after cannulating pulmonary artery, trachea tying right bronchus 10–15 days without any tumor cells as control group or with NSCLC (A549, H1299 H460), SCLC (H69, H446 SHP77) breast cell lines (MCF7 MDAMB231) through trachea. performed lobectomy, H&E staining IHC human mitochondria primary growth formation lesions. addition, isolated circulating (CTC) seeded GFP tagged cells. group, no gross nodules were found, showed hyperplastic mitochondria. All models formed that had microscopic characteristics on showing CTC those labeled they viable culture. Finally, all lesions stained shows can form tumor, an environment. This study suggests natural matrix scaffold only necessary component drive progression play role modulating progression.
