K-ras and p53 Gene Mutations in Pancreatic Cancer: Ductal and Nonductal Tumors Progress through Different Genetic Lesions

摘要: We studied K-ras and p53 gene mutations in a panel of 57 primary pancreatic cancers including ductal nonductal tumors. DNAs were obtained from formalin-fixed, paraffin-embedded material. Target sequences amplified by polymerase chain reaction analyzed denaturing gradient gel electrophoresis sequencing. Both genes frequently mutated (25 35, 71.4%; 18 51.4%, respectively). confined to the second position codon 12 where base transitions transversions equally observed. changes mainly missense mutations. Transitions found with prevalence G:C-->A:T among transitions. No alterations present 6 exocrine tumors (with one exception) endocrine analyzed.
