摘要: Cultured primary porcine urinary bladder epithelial cells (PUBEC) represent an adequate and easy to handle in vitro system for studies of urothelial toxicity. PUBEC maintain vivo-like metabolic activities physiological functions. They express inducible cytochrome P4501A isoenzymes, which are particular relevance, since they contribute activation carcinogens. A possible drawback is their isolation from common domestic pigs that do not inbred strain. In order further establish as a standard toxicity test we analysed interindividual differences CYP1A1 inducibility. Interestingly, observed by flow cytometry obtained individual consist two distinct subpopulations with non-inducible cells. strong, concentration-dependent induction was the responsive subpopulation when incubated benzo[a]pyrene (B[a]P) concentration range between 1 10 μM. contrast, no non-responsive up highest tested concentrations 100 The fraction showed large ranging 65% total cell number. For practical purposes it might be reasonable analyse pools five substantially reduce batch variability. conclusion, have identified functionally It will interesting study whether CYP1A subtype more susceptible
