摘要: The concept of sequential injection analysis is described, and an application this novel technique to assay traces a proteolytic enzyme demonstrated. automated method based on stopped-flow procedure with fluorescence detection. Instead using the conventional constant flow rate, sinusoldal pattern, as generated by robust cam-driven piston pump, has been applied. validation system nonreactive fluorescent compound yielded highly reproducible results over concentration range 3 order magnitude. calibration graph for activities in 5-75 micron mL-1 could be fitted second-order polynomial. detection limit 2.5 mL-1, which corresponds 7.2 ng pure active enzyme.
