摘要: The most reliable prognostic factors for patients with primary malignant brain tumors remain histology, age, and functional status. Management of these individuals might be improved by quantifying pertinent molecular markers. We have measured the gene dosage epidermal growth factor receptor (EGFR), mouse double minute 2 (MDM2), cyclin-dependent kinase 4 (CDK4) genes in a series tumor specimens correlated their amplification status standard survival. Individual DNA was successively hybridized probes EGFR, MDM2, CDK4. signal quantified densitometry, defined as > or = times normal. Survival, Karnofsky performance status, histology were amplification. Nineteen astrocytomas, 20 anaplastic 70 glioblastomas had complete data available. Median survival without any form 70.7 88.6 weeks, respectively (P 0.0369). For EGFR alone, those median 58.9 0.0104). By Cox analysis, only 0.04) 0.0157) significant independent predictors Gene itself not predictive survival, even 0.8249). lack correlation between may because CDK4 are portions larger signaling systems. Therefore, direct single outcome is entirely unexpected.
