Patterns of somatic mutation in human cancer genomes

摘要: Cancers arise owing to mutations in a subset of genes that confer growth advantage. The availability the human genome sequence led us propose systematic resequencing cancer genomes for would lead discovery many additional genes. Here we report more than 1,000 somatic found 274 megabases (Mb) DNA corresponding coding exons 518 protein kinase 210 diverse cancers. There was substantial variation number and pattern individual cancers reflecting different exposures, repair defects cellular origins. Most are likely be 'passengers' do not contribute oncogenesis. However, there evidence 'driver' contributing development studied approximately 120 Systematic sequencing therefore reveals evolutionary diversity implicates larger repertoire previously anticipated.
