摘要: OBJECTIVE: Previously, we demonstrated that the anti-epidermal growth factor receptor (EGFR) artibody cetuximab alone was effective against EGFR-amplified glioblastoma multiforme (GBM) cell in vivo and vitro. The purpose of present work to study further effectiveness as a monotherapy well combining it with cadiation therapy or chemotherapy. METHODS: GBM cells were implanted either flanks nude mice determine on larger tumor burden intracranially assess ability cross blood-brain barrier. Cells also exposed combination radiation chemotherapeutic agents vitro RESULTS: Increasing fumpr decreased amount inhibition. For first two intracranial models using for 5 weeks, treated had significant increase median survival compared controls. When given indefinitely, results encouraging, an group not yet reached but at least 900%. Mice flank than those treatment alone. Preliminary experiments showed increased cytotoxicity. CONCLUSION: These demonstrating GBM. Surprisingly, when admintered systemically intractanial model. Radiation augmented effect vivo. In analysis additive effects well. confirm EGFR blockade prtential treatrnent human
