摘要: Although several glioma models exist, systematic morphometrical studies on such experimental tumors are lacking. The purpose of this study was the quantitative assessment how rat strains, cell lines, injection techniques and location affect reproducibility histopathological features. Glioma cells were implanted in 3 brain locations, with different (free hand, stereotactic, water-tight device), variable volumes, concentrations infusion rates. Tumors developed from 2 lines (9L C6) immunocompetent (Wistar Fischer 344) immunodeficient rats (New Zealand). Animals underwent daily neurological examination. At scheduled time macro microscopically evaluated a analysis performed. C6 gliomas appeared very infiltrative irregularly shaped; 9L showed, by using same technique, grossly regular shape. Margins at tumor–brain interface macroscopically demarcated rats. In nude rats, more infiltrative, although regularly shaped, closer morphological resemblance to human gliomas. implantation frontal area, anterior nucleus caudatus (3 mm coronal suture) gave reproducible tumor shape size, no hydrocephalus early deterioration. use stereotactic technique or device, small volume (<10 µl) suspension, low rate useful reduce morbidity improve data reproducibility. No difference mortality observed model constitutes good option for reliable evaluation growth. We propose caudatus. This produced longest symptom-free survival.
