摘要: Mutations in codons 12 and 13 of the K-RAS oncogene are detected at a remarkably high frequency colorectal adenocarcinoma believed to be critical event oncogenesis. In present study, we evaluated tumor specimens from Taiwan for mutations using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis direct DNA sequencing. were found 48 181 (26.5%) tumors, 30 G→A transitions (62.5% all mutations), 14 G→T transversions (29.2%), only 4 G→C (8.3%). Similar relative mutation frequencies spectra regardless sex patient, grade, or stage. The among suggests that G/T mismatches play an important role oncogenesis adenocarcinoma, implying alkylating carcinogens may involved carcinogenesis. Although appears lower than those reported United States (40%), France (49%), Netherlands (34%), spectrum point gene Chinese population similar. reason these results diet ethnicity not rate limit factors controlling but influence on human adenocarcinomas.
