摘要: Abstract Worldwide used herbicide atrazine is linked to reproductive dysfunction in females. In this study, we investigated the effects and mechanism of action ovary using a primary culture immature granulosa cells. cells, follicle-stimulating hormone (FSH) activates both cyclic adenosine monophosphate (cAMP) extracellular-regulated kinase 1/2 (ERK1/2) cascades, with cAMP pathway being more important for luteinizing receptor (LHR) aromatase (CYP19A1) mRNA expression. We report that 48 h after exposure FSH-stimulated LHR CYP19A1 expression estradiol synthesis were decreased, sensitive than mRNA. Inadequate acquisition atrazine-exposed cells renders human chorionic gonadotropin (hCG) ineffective stimulate amphiregulin (Areg), epiregulin (Ereg), progesterone (Pgr) expression, suggesting anti-ovulatory effect atrazine. To dissect signaling cascade involved U0126, pharmacological inhibitor ERK1/2. U0126 prevents atrazine-induced decrease levels production ERK1/2 inactivation restores ability hCG induce ovulatory genes Cell-based ELISA assay revealed does not change phosphorylation The results from study reveal affect but requires cause FSH-induced thus compromising ovulation female fertility.
