Anti‐Cancer Drugs Reactivate Tumor Suppressor miR‐375 Expression in Tongue Cancer Cells

摘要: Head and neck cancer is one of the deadliest malignant diseases chemotherapy a common treatment option. Despite development chemotherapies for several decades, how these drugs affect dynamics gene regulation still largely unknown. In our previous study, miR-375 was shown to be underexpressed in oral cancers thus unable serve as tumor suppressor microRNA regulate certain putative oncogenes. this we found that anti-cancer reactivated tongue cells. Incubation cells CAL 27 SCC-25 medium containing doxorubicin, 5-fluorouracil, trichostatin A, or etoposide significantly increased expression its primary transcript pri-miR-375. The dose- time-dependent effects doxorubicin were demonstrated by increases response drug. Significant suppression pri-miR-375 observed human specimens decrease more prominent advanced stage tumors. Bioinformatics from four publicly available mRNA microarray data sets suggested candidate targets are mainly involved biology, indicating likely suppressed via due with drugs.
