摘要: Human ectopic pregnancy (EP) is a leading cause of pregnancy-related death, but the molecular basis underlying onset tubal EP largely unknown. Female Dicer1 conditional knockout mice are infertile with dysfunctional Fallopian tube and have different miRNA expression profile compared to wild-type mice, we speculated that Dicer-mediated regulation specific miRNA-controlled targets might contribute EP. In present study, used microarray analysis quantitative RT-PCR examine miRNAs core regulatory components in tissues from women We found levels DICER1, four (let-7i, miR-149, miR-182, miR-424), estrogen receptor α distinguished implantation site non-implantation site. Computational algorithms screening for interactions progesterone signaling pathways showed were predicted target ten genes, including NEDD4, TAF15, SPEN. Subsequent experiments differences NEDD4 mRNA protein between sites. Finally, revealed increases smooth muscle cell stromal parallel decrease epithelial SPEN, associated implantation.
