摘要: In diffuse gastric carcinoma, despite common E-cadherin gene (CDH1) mutations, tumors show absence of CDH1 loss heterozigosity (LOH) in most cases. This observation challenges the classical two-hit model tumor suppressor inactivation. order to investigate whether or not promoter methylation may function as second hit we analysed a series 23 sporadic carcinomas for presence methylation, LOH and expression. mutations were detected nine 16 (56.3%) none seven intestinal carcinomas. harboring was observed single case. Loss plasma membrane expression consistently found all cases with mutation, suggesting that inactivated remaining allele via different mechanism. diffuse-type carcinoma cases, including six (66.7%) mutations. also seen two (28.6%) intestinal-type Our results is more than half
