Inhibitory effect of curcumin on invasion of skin squamous cell carcinoma A431 cells.

摘要: OBJECTIVE To investigate the regulatory effect of curcumin on expression signal transducer and activator transcription 3 (STAT3) in skin squamous cell carcinoma tissues as well possible mechanisms prevention treatment carcinoma. MATERIALS AND METHODS Highly invasive A431 cells were treated with at various doses .The cytotoxic effects 5, 10, 15, 20, 25, 30, 35, 40 50 umol/L for 24, 48 72 hours measured by MTT assay. The invasion capacity 10 15 was Transwell test, while adhesive ability assessed adhesion 5,10 levels STAT3 determined Western blotting mRNA RT-PCR. RESULTS Treatment a more than 24 hour inhibited growth time-and dose-dependent fashion (p<0.001). less showed no significant cells, survival rates being 85%.The abilities decreased gradually increasing concentration, exerting strongest inhibitory (p<0.05). Curcumin level CONCLUSIONS may reduce inhibiting activation pathway target gene pathway.
