摘要: Endometrial cancer is characterized by alterations in the stromal cells and supporting extracellular matrix addition to intrinsic of malignant epithelial cells. We have developed a cell culture model that demonstrates role regulation proliferation, hormone responsiveness, differentiation an endometrial adenocarcinoma line (Ishikawa). Conditioned medium (CM) was collected from normal primary human grown on plastic or within basement membrane extract, Matrigel. The CM produced cultured contact with Matrigel markedly inhibited Ishikawa proliferation compared plastic. varied steroid treatment presence embedded When were placed coculture Matrigel, production differentiated secretory product, glycodelin, induced. Gene expression receptors, growth factors, integrins analyzed reverse transcription-PCR determine potential factors involved regulatory function. These combined studies imply phenotype can be induced differentiate more closely resemble epithelium reintroduction appropriate matrix. Additionally, study shows proteins influence function as they mediate growth.
