Stimulation of the Na(+)-K(+) pump in cultured guinea pig airway smooth muscle cells by serotonin.

摘要: The effect of 5-hydroxytryptamine (5-HT) or serotonin on Na + -K pump activity airway smooth muscle was investigated by measuring 86 Rb uptake in cultured guinea pig tracheal cells. consisted three distinct components, one sensitive to ouabain, bumetanide, and insensitive either inhibitor. 5-HT induced a concentration-dependent increase ouabain-sensitive (EC 50 = 21 nM) but had no bumetanide-sensitive uptake, suggesting that it stimulates the not -Cl − cotransporter. Ouabain-sensitive also stimulated 2A/2C agonists 2,5-dimethoxy-4-iodoamphetamine α-methyl-5-HT, 1 agonist 5-carboxamidotryptamine, 1A/1B/2C 1-(3-chlorophenyl)piperazine, 3 1-(3-chlorophenyl)biguanide. 5-HT-stimulated inhibited 2A antagonists ketanserin spiperone, 1A antagonist NAN 190 Y25310. reducing extracellular concentration -H exchange inhibitors dimethylamiloride 5-( N -methyl- -isobutyl)-amiloride. These observations suggest via receptors mechanism dependent influx, possibly through exchanger. Because stimulation produces hyperpolarization, this may represent negative-feedback opposes contraction response 5-HT.
