摘要: In the adult mammalian brain, new neurons and glia are continuously generated but molecular factors regulating their differentiation lineage relationships largely unknown. We show that Ascl1, a bHLH (basic helix-loop-helix) transcription factor, transiently labels neuronal oligodendrocyte precursors in brain. Using vivo tracing with inducible Cre recombinase, we followed maturation of these four distinct regions. hippocampus, Ascl1 mostly marks type-2a progenitor cells some late stage type-1 stem cells. Thirty days after expression, although majority matured to granule neurons, few remained as immature progenitors. By 6 months, however, essentially all were neurons. contrast, olfactory bulb lineage, is restricted transit amplifying cells, by 30 d into GABAergic interneurons. also broadly subcortical gray white matter corpus callosum, defines ventral layer early do not yet express other markers this like PDGFRalpha Olig2. d, most had transitioned mature oligodendrocytes. expressing already coexpressed markers, they precursors. Our results reveal common marker progenitors both oligodendrocytes defined dynamics different brain
